Anti-5-Hydroxymethylcytosine (5hmC), clone EDL HMC 1A, Cat. No. MABE1093, is a highly specific mouse monoclonal antibody that targets DNA with 5-hydroxymethylcytosine (5hmC) modification and has been tested in Dot Blot and Immunoprecipitation.
Dot Blot Analysis: 0.5 µg/mL from a representative lot detected 0.39-100 ng of DNA with 5-hydroxymethylcytosine (5hmC) modification, but not DNA with only 5-methylcytosine (5mC), 5-formylcytosine (5fC), 5-carboxylcytosine (5caC) modification or unmodified cytosine (Courtesy of Jeong-Heon Lee, Ph.D.; Chad Clark; Tamas Ordog, M.D.; Zhiguo Zhang, Ph.D. Epigenomics Development Laboratory, Epigenomics Program, Center for Individualized Medicine, Mayo Clinic, Rochester, Minnesota, USA).

General description
5-Hydroxymethylcytosine (also known as 5-hmC) is the first oxidative product in the active demethylation of 5-methylcytosine (5mC). 5hmC is a DNA pyrimidine nitrogen base and is a predominantly stable DNA modification. It is produced by the action of three Ten-eleven translocation 1 (TET1) enzyme on 5mC. Each mammalian cell contains 5hmC, but its levels vary significantly depending on the cell type. Higher levels of 5hmC are found in neuronal cells and its level is shown to increase with age in hippocampus and cerebellum. It is also prevalent in embryonic stems cells and its reduced levels are shown to result in impaired self-renewal of stem cells. In the brain and in embryonic stem cells, 5hmC in enriched in promoters, gene bodies, and intergenic areas near genes and positively correlates with gene expression at these loci. 5hmC appears to play a role to promote gene expression during active demethylation. Significantly lower of 5hmC have been reported in cancer cell lines, melanoma, adenomas, and carcinomas compared to the normal surrounding tissue.
